Design Notes

Synthesis of whole gene from IDT was not possible due to high complexity score of the sequence. The sequence was fragmented into four smaller fragments using NEB Assembly Tool. The fragments and the primers for PCR are available as separate parts. Gene was codon optimized for BL21 and K12 using IDT and GenScript's tool. We were able to get the gene synthesized flanked by appropriate restriction enzymes for cloning through GenScript.

Scl2.28-1 allele comes from the cell surface of group A Streptococcus(GAS), a Gram-positive bacterium, specifically serotype M28 strain MGAS6274.
